Victoria Police requested the assistance of MFB firefighters in Bundoora in the early hours of Saturday morning.
Crews arrived on scene just after 3am and were met with a fire that had taken hold of the second floor and was extending to the roof of a home on May Street.
Wearing breathing apparatus for their safety, firefighters entered the property to conduct a search and ensure all occupants had evacuated, and began to attack the fire.
Three occupants were transported to hospital in the care of Ambulance Victoria paramedics.
Approximately 30 firefighters were able to get the fire under control within half an hour of their arrival on scene.
The cause of the fire is believed to be suspicious, and it will be investigated by Victoria Police.
At the height of the fire there were six MFB firefighting appliances on scene including an aerial teleboom appliance, as well as four other support and Command units.
